In vitro | CCR7 Ligand 1 (CCR7-Cmp2105) specifically targets and binds to a distinct site within the intracellular domain of CCR7, located between the terminal regions of transmembrane segments TM1, TM2, TM3, and TM6, as well as the loop linking TM7 and helix 8 [1]. This interaction allosterically impedes the attachment of the natural chemokine CCL19 to CCR7, evidenced by an inhibition concentration (IC50) of 35 nM in scintillation proximity assays (bottom) [1]. Additionally, the application of CCR7 Ligand 1 at varying concentrations (20, 40, 60, 80, 100 μM) demonstrates a concentration-dependent enhancement of CCR7's thermal stability [1]. |
