In vivo | In an experimental setting using male Sprague Dawley rats weighing 150-175 g (NTG 10 mg/kg; i.p.), AKU-005 administered at a dosage of 0.5 mg/kg intraperitoneally demonstrated both antinociceptive and anti-inflammatory effects. Administered 3 hours after nitroglycerin (NTG), AKU-005 significantly reduced NTG-induced facial rubbing behaviors and downregulated CGRP mRNA levels in the meninges, medulla, CSC, and ipsilateral TG. Additionally, it effectively inhibited the rise in IL-6 and TNF-alpha mRNA in all studied regions, with the exception of TNF-alpha in the medulla. |
