Description
AKU-005 is a dual inhibitor of FAAH and MAGL, exhibiting IC50 values of 63 nM and 389 nM against rat and human FAAH, respectively. This compound has potential for researching trigeminal nociceptive hypersensitivity.
In vivo
In an experimental setting using male Sprague Dawley rats weighing 150-175 g (NTG 10 mg/kg; i.p.), AKU-005 administered at a dosage of 0.5 mg/kg intraperitoneally demonstrated both antinociceptive and anti-inflammatory effects. Administered 3 hours after nitroglycerin (NTG), AKU-005 significantly reduced NTG-induced facial rubbing behaviors and downregulated CGRP mRNA levels in the meninges, medulla, CSC, and ipsilateral TG. Additionally, it effectively inhibited the rise in IL-6 and TNF-alpha mRNA in all studied regions, with the exception of TNF-alpha in the medulla.
