Description
P163-0892 is a highly effective and specific antifungal compound targeting Cryptococcus species. Additionally, P163-0892 demonstrates moderate capability to penetrate the blood-brain barrier [1].
In vitro
P163-0892, at concentrations ranging from 0-32 μg/mL over 48 hours, demonstrates antifungal efficacy, achieving minimum inhibitory concentrations (MICs) of 0.25 μg/mL and 0.5 μg/mL against Cryptococcus neoformans and Cryptococcus gattii, respectively. Notably, it also exhibits favorable aqueous solubility and is devoid of cytotoxic and significant cardiotoxic effects.
In vivo
P163-0892 (10 mg/kg; 5 days) significantly extends the survival of larvae infected with either C. neoformans or C. gattii [1]. Animal Model: Wax moth larva infected with C. neoformans H99 or C. gattii WM178 [1]. Dosage: 10 mg/kg. Administration: 5 days. Result: Increased survival rate of larvae. Animal Model: Male Sprague–Dawley rats (200–250 g) [1]. Dosage: 2 mg/kg or 5 mg/kg. Administration: Intravenous or oral (Pharmacokinetic Analysis). Result: Pharmacokinetic Parameters of P163-0892: iv 2 mg/kg: C0 2460 ng/mL, Cmax 7.61 ng/mL, T1/2 749 h, Tmax 172 h, AUC0-inf 2.63 (h ng/mL), Cl 7.12 (mL/h/kg), MRT0-t 3.17 h, Vdss (L/kg) 7.12, BA (%) po 5 mg/kg: C0 2.97 ng/mL, Cmax 15.3 ng/mL, T1/2 3.17 h, NR.
