Compound B12 (PHGDH-IN-5) acts as a covalent inhibitor of PHGDH, demonstrating an IC50 value of 0.29 μM. It has been shown to inhibit the proliferation of cancer cell lines that overexpress PHGDH and reduce the production of glucose-derived serine in MDA-MB-468 cells [1].
In vitro
PHGDH-IN-5 (Compound B12) inhibits the proliferation of MDA-MB-468, NCI-H1975, HT1080, and PC9 cells with IC₅₀ values ranging from 9.93 μM to 21.2 μM [1]. At concentrations of 10-30 μM, PHGDH-IN-5 reduces the production of M+3-serine derived from 13C6-glucose in MDA-MB-468 cells [1].
