In vitro | Pilsicainide (SUN 1165 free acid; 10-200 μg/mL) dose-dependently reduces the peak amplitude of the net inward current, with an IC50 of 29.2 μg/mL in levo-thyroxine (T4)-treated rat atrial cells [1]. |
In vivo | Pilsicainide (SUN 1165 free acid; 2 mg/kg; i.v.; once) significantly reduces conduction velocity in the atrium of T4-treated male Sprague-Dawley rats (weighing 200-220 g) by decreasing Max dV/dt and net inward current, and altering electrocardiographic parameters. Administered as a bolus into the right external carotid vein over two minutes, pilsicainide extends the QT interval at 15 and 60 minutes post-administration, decreases the durations of P wave and QRS complex, reduces action potential amplitudes (APA) and Max dV/dt, and lengthens action potential duration. |
