Description
UBE2T/FANCL-IN-1 is a potent inhibitor of UBE2T/FANCL-mediated FANCD2 monoubiquitylation, significantly enhancing cellular sensitivity to Carboplatin, a DNA cross-linking agent.
In vitro
UBE2T/FANCL-IN-1 (CU2) effectively prevents GST-FANCLRING ubiquitylation at 100 μM and significantly reduces FANCD2 ubiquitylation mediated by HU and Cisplatin (10 μM) at 500 μM over 6, 12, and 24 hours. Additionally, 250 μM UBE2T/FANCL-IN-1 combined with 15 μM carboplatin markedly inhibits cell proliferation, resulting in less than 50% confluence after 8 days[1].
