Description
Anticancer agent 71 (Compound 4b) is a potent anticancer agent that arrests the cell cycle at the G2/M phase and induces apoptosis by upregulating Bax, Ikb-α, and cleaved PARP, while downregulating Bcl-2 expression levels.
In vitro
Anticancer agent 71 (Compound 4b), administered at concentrations ranging from 0.1 to 50 μM for 24 hours, significantly enhances cytotoxic effects and demonstrates robust antitumor potential in SW480 and PC3 cells [1]. This compound, at a specific dose of 11.08 μM applied to SW480 cells for the same duration, notably induces G2/M phase arrest [1] and triggers apoptosis. The apoptosis mechanism involves the upregulation of Bax and Ikb-α, along with cleaved PARP, and the downregulation of Bcl-2 expression [1]. Through cell cytotoxicity assays, it was determined that agent 71 inhibits cell viability with IC 50 values of 11.08 μM for SW480 cells and 17.39 μM for PC3 cells. Additionally, cell cycle analysis revealed a 45.3% increase in the G2/M phase population in SW480 cells. Apoptosis analysis recorded early and late apoptosis rates at 5.69% and 3.52%, respectively. Western blot analysis confirmed these findings by showing a decrease in Bcl-2 expression and an increase in Bax expression, highlighting the compound's ability to effectively promote apoptosis in cancer cells [1].

In Vivo Formulation Calculator (Clear solution)

Please enter your animal experiment information in the following box and click Calculate to obtain the mother liquor preparation method and in vivo formula preparation method:
TargetMol | Animal experimentsFor example, your dosage is 10 mg/kg Each animal weighs 20 g, and the dosage volume is 100 μL . TargetMol | Animal experiments A total of 10 animals were administered, and the formula you used is 5% TargetMol | reagent DMSO+30% PEG300+5% Tween 80+60% ddH2O. So your working solution concentration is 2 mg/mL。
Mother liquor preparation method: 2 mg of drug dissolved in 50 μL DMSOTargetMol | reagent (mother liquor concentration of 40 mg/mL), if you need to configure a concentration that exceeds the solubility of the product, please contact us first.
Preparation method for in vivo formula: Take 50 μL DMSOTargetMol | reagent main solution, add 300 μLPEG300TargetMol | reagent mix well and clarify, then add 50 more μL Tween 80, mix well and clarify, then add 600 more μLddH2OTargetMol | reagent mix well and clarify
