Description
Acloproxalap is a quinoline-based aldehyde scavenger used in studies of diseases with toxic aldehyde accumulation, including inflammatory diseases of the eye and skin, respiratory diseases such as pneumonia, organ diseases, and syndromes associated with viral infections[1] [2].
In vivo
Acloproxalap (compound I-6) administered at doses of 100 or 200 mg/kg via intraperitoneal injection or oral gavage daily for 6 days has been shown to significantly alleviate colon inflammation in acute ulcerative colitis (UC) in female Swiss Webster mice, according to study [1]. Administration specifics include intraperitoneal injections at 100 mg/kg and oral gavage at 200 mg/kg, demonstrating notable inhibition of weight loss in the former without significant differences in the latter when compared to control groups. Both methods significantly ameliorated colonic histopathology. In another study involving male non-naïve beagle dogs [2], a single intravenous injection of Acloproxalap (compound I-1) at 3 mg/kg revealed pharmacokinetic parameters that included a half-life (t 1/2) of 1.82 hours and a clearance rate of 12.2 mL/kg/min. Oral administration at 10 mg/kg showcased a longer half-life (3.02 hours), maximum concentration (C max) of 3558 ng/mL, and a bioavailability (BioA) of 75%.
