In vitro | FDGlcU, at a concentration of 0.5 μg/mL over 12 hours, demonstrates enzymatic activity and inhibits βG activity in live bacteria [1]. |
In vivo | FDGlcU (7.3 μM/kg) is used for in vivo time-lapse imaging of bacterial βG activity [1]. For whole-body imaging of intestinal βG activity [1], administer 50 μL of DDW, FDGlcU (7.3 μM/kg), and fluorescein (7.3 μM/kg) via gavage on the day of imaging. Anesthetize the mice with isoflurane and perform whole-body optical imaging using an IVIS spectrum optical imaging system with a GFP filter set. In a biodistribution study, sacrifice the mice 3 hours after gavage with FDGlcU or fluorescein (λex = 480 nm, λem = 514 nm). This protocol should be modified according to specific needs. |
