In vitro
GABAA receptor agent 7 (compound 5c) exhibits anticonvulsant properties, demonstrating an inhibitory concentration (IC50) of 0.452 μM in the 4-AP induced hyper-excitability model [1]. Additionally, it enhances GABA-induced activation of the GABAA1 receptor dose-dependently within a concentration range of 0.01-100 μM over a period of 0-140 seconds, with an effective concentration (EC50) of 3.08 μM [1].
In vivo
GABAA receptor agent 7 demonstrates anticonvulsant properties in mice, achieving an effective dose 50 (ED50) of 31.81 mg/kg and a toxic dose 50 (TD50) of 547.89 mg/kg [1]. This compound specifically mitigates seizures induced by pentylenetetrazol (PTZ) without affecting those caused by maximal electroshock (MES) in mice [1].
