Description
hMAO-B/MB-COMT-IN-2 is a dual inhibitor of MAO-B/MB-COMT with IC 50s of 4.27 μΜ and 2.69 μΜ for hMAO-B and MB-COMT, respectively. hMAO-B/MB-COMT-IN-2 protects cells from oxidative damage that can be used in the neurodegeneration disease research, for example, Parkinson’s Disease (PD) [1].
In vitro
hMAO-B/MB-COMT-IN-2 (compound 8) demonstrates various effects on differentiated SH-SY5Y cells; at a concentration of 50 μM over 24 hours, it reduces resazurin reduction and induces lysosomal toxicity through reactive oxygen species (ROS) production. Contrastingly, at 10 μM for 30 minutes, it exhibits significant cytoprotective properties against t-BHP. Moreover, it is capable of crossing the blood-brain barrier (BBB) via passive diffusion, as determined by the PAMPA-BBB kit. Additionally, a concentration range of 30-50 μM over 24 hours has been confirmed to have a good safety profile in SK-N-SH cells [1].
