In vitro | Human PTHrP-(1-36), with an EC50 of 0.05 nM, augments intracellular calcium levels in human epidermal keratinocytes [2]. At a concentration of 100 nM over 24 hours, it stimulates proliferation in human β-cells [3]. Similarly, when applied at 100 nM for 30 minutes, PTHrP-(1-36) boosts insulin secretion in human islets [3]. In mice, PTHrP-(1-36) elicits a swift Ca2+ response in UMR 106 cells at an EC50 of 1 nM [4]. |
In vivo | PTHrP-(1-36) (mouse, 160 μg/kg, subcutaneously, 5 days weekly for 7, 30, or 90 days) promotes beta cell regeneration and augments beta cell mass following partial pancreatectomy in mice [5]. Additionally, PTHrP-(1-36) (mouse, 100 μg/kg, subcutaneously, administered biweekly) counteracts the reduced Wisp1 expression in diabetic mouse models [6]. |
