Description
Kelatorphan is an enkephalin degrading enzyme full inhibitor.
In vivo
Kelatorphan (20 mg/kg i.v.) increases ventilation in both arthritic and non-arthritic rats. In pentobarbital-anesthetized rats, Kelatorphan (20 mg/kg, n=6) significantly elevates respiration by 116% within 10±15 minutes post-administration. In normal awake rats, Kelatorphan (10±20 mg/kg i.v.) enhances minute-volume through a dose-dependent increase in breathing frequency. Kelatorphan alone (50 μg) significantly boosts intact [3H]enkephalin content, corresponding to 80±11% of total recovered radioactivity[2]. Naloxone (0.2 mg/kg i.v.) pre-treatment does not antagonize Kelatorphan, but a larger dose (1 mg/kg i.v.) significantly counteracts Kelatorphan (20 mg/kg) at 5 and 10 minutes in awake rats[3].
