Description
M62812 (free base), a Toll-like receptor 4 (TLR4) signal transduction inhibitor, suppresses activation of endothelial cells and leukocytes and prevents lethal septic shock in mice. It is utilized in sepsis research [1].
In vitro
M62812 (0-10 μg/ml) suppresses the LPS-induced upregulation of inflammatory cytokines, adhesion molecules, and procoagulant activity in human endothelial cells and peripheral mononuclear cells [1].
In vivo
M62812, administered intravenously at dosages of 10-20 mg/kg as a single injection, protected mice from lethality in a d-galactosamine-sensitized endotoxin shock model, also reducing inflammatory and coagulation parameters [1]. Moreover, a daily dose of 20 mg/kg of M62812 prevented mortality in mice subjected to a cecal ligation and puncture model [1].
