Description | NSC 80467, a selective DNA damaging agent, functions by inhibiting survivin. Specifically, it preferentially targets DNA synthesis, leading to the induction of two distinct markers of DNA damage, namely γH2AX and pKAP1. |
In vitro | NSC 80467 exhibits an activity profile against the NCI-60 cell line panel similar to its suppression of survivin expression and correlates with DNA-damaging agents through COMPARE analysis. It preferentially targets DNA synthesis and triggers a DNA damage response, marked by increased levels of γH2AX and pKAP1, at doses lower than those required to suppress survivin expression. At concentrations of 200 or 800 nM over 24 hours, NSC 80467 effectively inhibits survivin expression in PC3 cells, as demonstrated by Western Blot Analysis[1]. |
