NW16 is an orally active STAT3 inhibitor with a Kd of 11.0 μM. It causes cell cycle arrest at the S phase, induces apoptosis in HCT116 cells, and inhibits the proliferation of cancer cells HCT116, A549, and B16, with IC50 values of 0.28 μM, 1.22 μM, and 9.86 μM, respectively. NW16 promotes the production of ROS and inhibits the PI3K/AKT signaling pathway, demonstrating antitumor activity in mouse models.
