Bioactivity
Description
PNC-28, a peptide derived from the mdm-2-binding domain of p53 protein (residues 17–26), incorporates a membrane-crossing penetratin sequence. This compound is utilized in pancreatic cancer research [1] [2].
In vitro
PNC-28 demonstrates activity by selectively binding to and penetrating cancer cell membranes, inducing apoptosis. This effect is notably absent in normal, non-cancerous cells due to the specific interaction with a distinct motif on the cancer cell surface. Additionally, PNC-28 facilitates the inhibition of tumor growth and metastasis.
In vivo
PNC-28, administered subcutaneously (SC) or intraperitoneally (IP) at a dose of 2 mg/mouse for 14 days, effectively impedes the growth of BMRPA1. Tuc3 cells in vivo [1]. Additionally, doses ranging from 1 to 20 mg/mouse, when given subcutaneously for the same duration, notably restrain BMRPA1. Tuc3 tumor proliferation, with pronounced efficacy when applied directly to the neoplasm [1].
