In vitro | In isolated heart preparations from guinea-pigs, the compound Ro 363 induced arrhythmic contractions when administered at doses that elicited 70-100% of its maximal heart rate-increasing effects [1]. Additionally, in tracheal tissues of guinea-pigs that contract spontaneously, Ro 363 acted as a potent full agonist, demonstrating approximately half the effectiveness of (-)-Isoprenaline. This activity of Ro 363 is attributed to its activation of β1-receptors, as evidenced by its comparable potency to (-)-Isoprenaline in affecting the trachea, heart, and ileum [2]. |
In vivo | In chloralose-anesthetized felines, Ro 363 exhibits negligible arrhythmogenic activity compared to epinephrine (adrenaline), especially in subjects whose cardiac sensitization has been induced by either U-0882 or halothane [1]. |
