In vitro | The dopamine D1 receptor protein presence was confirmed in the native human airway epithelium and in three types of cultured human airway epithelial cells. Treatment with A68930 hydrochloride (1 μM) across varying durations (5-60 minutes for 16HBE14o- or NCI-H292 cells; 48 hours for NCI-H292 cells) demonstrated significant biochemical changes. Notably, a brief exposure (1 μM; 5-60 minutes) in 16HBE14o- or NCI-H292 cells markedly enhanced the phosphorylation of cAMP response element-binding (CREB) protein, as evidenced by Western Blot Analysis. Prolonged exposure (1 μM; 48 hours) in NCI-H292 cells was found to induce MUC5AC mRNA expression, simultaneously boosting both the mRNA levels and protein expression of MUC5AC, confirmed through RT-PCR and Immunofluorescence techniques. Furthermore, a short exposure (1 μM; 20 minutes) in NCI-H292 cells significantly increased intracellular cAMP levels, indicating the compound’s efficacy in activating intracellular signaling pathways critical in respiratory epithelial cell functions [1]. |
