In vitro | The most common application of Ribonuclease A is to remove RNA during plasmid DNA or genomic DNA preparation. Preparation of Ribonuclease A Stock Solution:Prepare a 10 mg/mL Ribonuclease A stock solution using 10 mM sodium acetate (pH 5.2).Heat at 100°C for 15 minutes. Cool to room temperature, add 1/10 (v/v) 1 M Tris-HCl (pH 7.4), and adjust the pH to 7.4.Store at -20°C for stable storage for up to 2 years. Note: Boiling the Ribonuclease A solution under neutral conditions may lead to the precipitation of RNase. Boiling at a lower pH, if precipitation occurs, may be due to the presence of protein impurities. In such cases, the precipitate can be removed by high-speed centrifugation (13,000 rpm). |
