In vivo | Trichlormethiazide, administered at 1 mg/kg orally once, was observed to elevate urinary volume along with sodium and potassium excretion in rats. A higher dose of 10 mg/kg, administered intravenously every day for 5 days, substantially decreased mean arterial pressure (MAP) within 24 hours in male Sprague-Dawley rats (weighing 350-450 g) with high salt intake receiving angiotensin II, without affecting MAP in other groups. This compound showed pronounced effects in increasing potassium excretion in normal male Wistar rats (weighing 170-300 g) and enhanced urinary volume, sodium, and potassium excretion in rats with cisplatin-induced acute renal failure (ARF). Conversely, when administered at 10 mg/kg intravenously daily for 15 days, it effectively lowered MAP in rats subjected to a combination of angiotensin II and high salt intake, indicating its potential benefits in managing hypertension under specific dietary conditions. |
