Nemvaleukin alfa (ALKS 4230) is an interleukin 2 fusion protein, a selective agonist of the medium-affinity IL-2 receptor, produced by fusion of the IL-2Ralpha subunit of the circularly arranged interleukin-2 (IL-2) and IL-2 receptor (IL-2R) complexes, which drives antitumor Immunity and inhibits tumor growth in small cell lung cancer.

In vitro | Nemvaleukin alfa was able to activate lymphocyte subpopulations in cynomolgus monkeys and human samples with EC50 values ranging from 0.45 to 2.2 nM. Nemvaleukin alfa induced the activation of medium-affinity IL-2R on human cells (HH cells) after treatment for 30 minutes at concentrations ranging from 0-100 nM. Under the same conditions, Nemvaleukin alfa was also able to induce the expression of pSTAT5 in NK cells with an EC50 value of 0.45 nM. In addition, Nemvaleukin alfa was able to induce the activation of effector lymphocytes isolated from human peripheral blood when treated at a concentration of 0.5 nM for 5 days. [2][3] |
In vivo | In a mouse model, Nemvaleukin alfa significantly inhibited small cell lung cancer (SCLC) tumor growth when administered subcutaneously at a dose of 6 mg/kg every 4 days.[1] |
