Description
PNU288034, a potent oxazolidinone antibiotic, demonstrates antimicrobial activity for the prevention and treatment of Gram-negative infections.
In vitro
In vitro studies, conducted to characterize the involved transporters, demonstrate uptake of PNU-288034 by human organic anion transporter 3 (OAT3; Km = 44 +/- 5 μM) and human multidrug and toxin extrusion protein 1 (hMATE1; Km = 340 +/- 55 μM)[1].
In vivo
In liver microsomes across species, PNU-288034 exhibits metabolic stability, and the unchanged drug is cleared in the urine through an apparent active renal secretion process in rat and monkey. Coadministration of PNU-288034 with the OAT3 inhibitor probenecid significantly increases the plasma area under the curve (170%) and reduces both plasma and renal clearance in monkey. Furthermore, coadministration of PNU-288034 with cimetidine also reduces plasma clearance in rats to a rate comparable to probenecid coadministration[1].
