Description
Maleimido-tri(ethylene glycol)-propionic acid is a cleavable ADC linker used in the synthesis of antibody-drug conjugates (ADCs) for the preparation of neolymphostin-based ADC precursors, specifically for site-specific cysteine mutant trastuzumab-A114C conjugation[1].
In vitro
Mal-PEG3-C2-acid is utilized in the synthesis of linker-payload compounds 16 (compound 15) and 20 (compound 18), which are then conjugated to DAR1.9 and DAR1.7, respectively[1]. ADCs 23 and 24, which are Neolymphostin ADCs, incorporate DAG 1.9 and DAG1.7, respectively, linked to PIKK inhibitors through linker-payloads 16 and 20. Notably, ADC24 exhibits cytotoxic activities against BT474 and N87 cell lines with IC50 values of 195 and 202 nM, respectively[1]. Similarly, ADC23 shows cytotoxic effects on the same cell lines, with IC50 values of 286 and 274 nM, respectively[1].
