In vivo | Intravenous injection of 3 mg/kg of E-0747 through a femoral vein cannula over a period of seconds using four canine models of ventricular arrhythmia: digitalis, sedation, arrhythmia, and heart failure concluded that E-0747 inhibits arrhythmia by inhibiting Na channels in cardiomyocytes[1];At a frequency of 1 Hz and a concentration higher than 3 X 10(-7) M, E-0747 decreased the maximum rate of rise (Vmax) of the action potential of Purkinje fibers in porcine heart, whereas the resting membrane potential was unchanged, and E-0747 inhibited Vmax in a rate-dependent manner.The inhibition was about 50% at a concentration of 3 X 10(-6) M with a stimulus of 3.3 Hz.[4]. |
