Description
IPI-493 (17-amino17demethoxy) is a novel HSP90 inhibitor with antitumor activity.
In vivo
In Nude mice of GIST-BOE, the oral doses of IPI-493 were 50 mg/kg twice daily, 40 mg/kg once daily, and 100 mg/kg three times weekly. In the GIST-PSW and GIST-48 models, the dose of IPI-493 was reduced to 80 mg/kg with no change in the duration of dosing.IPI-493 treatment resulted in tumor growth stabilization, different degrees of proliferation inhibition, induction of apoptosis and necrosis, and down-regulation of the KIT and its signaling cascade, especially in the GIST-BOE model.IPI-493 treatment significantly reduced vascular density.[5]
