A novel and highly conserved binding site at the interface of the protease and helicase domains of the Hepatitis C Virus (HCV) NS3 protein has been identified. Binding of 4-Phenoxybenzylamine to this allosteric site inhibits NS3 function by stabilizing an inactive conformation, introducing a new class of direct-acting antiviral agents.
