In vitro | The potent compound (SBC-115,337) showed an IC50 of 0.6 μM measured in an in vitro ELISA assay to monitor the effect of PCSK9 binding to the recombinant LDLR. All the investigated compounds exhibited no effect on the synthesis, processing and secretion of PCSK9. Compound SBC-115,337 at 1.2 μM induced more than tenfold upregulation of LDLR in HepG2 cells with respect to the control, increased uptake of fluorescently labeled DiI-LDL and lowered LDL-c levels in mice fed a high-fat diet.[1] |
