In vitro | Ropinirole has a 10- to 20-fold higher binding affinity for D3 receptors than for D2 and D4 receptors. As an orally potent D3/D2 receptor agonist, Ropinirole has a Ki value of 29 nM for the D2 receptor and has pEC50s of 7.4, 8.4, and 6.8 for the hD2, hD3, and hD4 receptors, respectively, with no affinity for the D1 receptor. [1] Ropinirole showed low activity at α2-adrenergic receptors and 5-HT2 receptors, but no activity at 5-HT1 receptors, benzodiazepine receptors, γ-aminobutyric acid receptors, and α1 and β-adrenergic receptors. In vivo | Intraperitoneal injections of Ropinirole at doses of 0.1-10 mg/kg were able to lower the threshold for intracranial self-stimulation (ICSS), showing anxiolytic and antidepressant effects without affecting motor function or spatial memory. [2] |
