Penciclovir (BRL 39123) is a Herpesvirus Nucleoside Analog DNA Polymerase Inhibitor. In HSV infected cells, penciclovir is phosphorylated by viral thymidine kinase and subsequently converted by cellular kinases into the active metabolite, penciclovir triphosphate, which competitively inhibits viral HSV polymerase by blocking deoxyguanosine triphosphate substrate binding. As a result, herpes viral DNA synthesis and replication are selectively inhibited.

Tivicilovir (AM188) is a hepatitis B virus inhibitor. tiviclovir is a guanine-related acyclic 2'-deoxyguanine analogue with anti-herpesvirus activity.

Cyclopropavir (Filociclovir; MBX-400) is a compound of broad-spectrum anti-herpesvirus. It has good antiviral activity against cytomegalovirus (CMV), herpes simplex virus (HHV)-6 and HHV-8 (EC50s: 0.7 μM to 8 μM).

Ganciclovir (BW 759) hydrate is a nucleoside analogue that is an orally active antiviral agent with anti-cytomegalovirus activity. Ganciclovir hydrate is also active against members of the herpes group and some other DNA viruses in vitro. Ganciclovir hydrate inhibits the replication of human herpes viruses (HSV 1 and 2, CMV) and adenovirus serotypes 1, 2, 4, 6, 8, 10, 19, 22 and 28 in vitro. Ganciclovir hydrate also has an IC 50 of 5.2 μM against feline herpesvirus type-1 ( FHV-1 ) and can diffuse into the brain [1] [2] [3].
