Anaphase Promoting Complex/Cyclosome (APC/C) is a multisubunit E3 ubiquitin ligase complex that regulates the progression of the cell cycle, particularly during mitosis and the G1 phase. APC/C targets key cell cycle regulators, such as Securin and Cyclin B, for ubiquitin-mediated proteasomal degradation, thereby promoting the transition from metaphase to anaphase and the exit from mitosis. It ensures proper chromosome segregation by triggering the activation of Separase, which cleaves cohesin complexes holding sister chromatids together. APC/C activity is tightly regulated by co-activators like Cdc20 and Cdh1, which confer substrate specificity at different stages of the cell cycle. Dysfunction of APC/C can lead to chromosomal instability and is associated with various diseases, including cancer.
