T-cell-specific surface glycoprotein CD28, contains an Ig-like V-type domain. CD28 is one of the proteins expressed on T cells that provide co-stimulatory signals, which are required for their activation. It is the receptor for CD80 and CD86. When activated by Toll-like receptor ligands, the CD80 expression is upregulated in antigen presenting cells (APCs). The CD86 expression on antigen presenting cells is constitutive. CD28 is the only B7 receptor constitutively expressed on naive T cells. It involved in T-cell activation, the induction of cell proliferation and cytokine production and promotion of T-cell survival.

Interleukin12 receptor subunit beta 1 (IL12RB1) is a type I transmembrane protein that belongs to the hemopoietin receptor superfamily. IL12RB1 can spontaneously form homodimers and -oligomers, which are able to bind IL12 with only low affinity. CD27 antigen is also known as CD27L receptor, T-cell activation antigen CD27, Tumor necrosis factor receptor superfamily member 7, T14 and TNFRSF7. In humans, it is encoded by the CD27 gene. CD27 is a single-pass type I membrane protein with 3 TNFR-Cys repeats. It is a member of the TNF-receptor superfamily and is required for generation and long-term maintenance of T cell immunity. It binds to ligand CD70, and plays a key role in regulating B-cell activation and immunoglobulin synthesis. It plays a role in survival of activated T-cells and apoptosis through association with SIVA1.

ILDR2 is a member of the B7-like family of proteins that regulate T cell activity, is also a known endoplasmic reticulum molecule that regulates lipid homeostasis. The human ILDR2 lumenal domain shares a 99% and 98% homology with the mouse and rat respectively. The human gene encoding ILDR2 is located in a region on Chr1q23–25 that has been associated with type 2 diabetes. ILDR2 plays critical roles in hepatic clearance of lipoproteins and in lipid homeostasis. ILDR2 regulates human dendritic cells (DC2 cells, a subpopulation of polarized DCs that promotes Th2 differentiation). Recent publications reported that ILDR2 displayed negative regulatory functions on human and mouse T cells in various experimental systems. Fusion protein of ILDR2 lumenal domain with an Fc fragment, displays therapeutic effects in collagen-induced arthritis (CIA), a mouse model of rheumatoid arthritis (RA). ILDR2 represents a novel B7-like ligand that exerts negative immune modulation via interaction with a putative counterpart receptor expressed on activated T cells.

Human IL-2 receptor alpha (IL-2 R alpha), also known as Tac antigen and as CD25, was initially identified as a 55 kDa membrane glycoprotein that is capable of binding IL‑2.eceptor for interleukin-2. The receptor is involved in the regulation of immune tolerance by controlling regulatory T cells (TREGs) activity. TREGs suppress the activation and expansion of autoreactive T-cells. SEMA4D is a member of the semaphorin family,contains one Ig-like C2-type domain, one PSI domain and one Sema domain. SEMA4D is strongly expressed in lymphoid tissues, especially in the thymus, as well as in the nervous tissues. However, SEMA4D is expressed at lower levels in testes, brain, kidney, small intestine, prostate, heart, placenta, lung and pancreas, but not in colon and liver. SEMA4D is a cell surface receptor for PLXN1B and PLXNB2 that plays an important role in cell-cell signaling. C-type lectin domain family 2, member D (CLEC2D) is implicated in the immune response. Pre-eclampsia and HIV infection have opposing immune responses. Killer cell immunoglobulin-like receptor 2DL4(KIR2DL4) is a Single-pass type I membrane protein and contains 2 Ig-like C2-type (immunoglobulin-like) domains.It belongs to the immunoglobulin superfamily. KIR2DL4 is expressed in all NK cells and some T cells. KIR2DL4 activates the cytotoxicity of NK cells, despite the presence of an immunoreceptor tyrosine-based inhibition motif (ITIM) in its cytoplasmic tail. The ITIM was not necessary for activation of lysis by KIR2DL4. The activation signal of KIR2DL4 was sensitive to inhibition by another ITIM-containing receptor. Interleukin-18 (IL-18) is a member of the interleukin-1 family of cytokines produced constitutively by different cell types and by adipose tissue. C-type lectin domain family 2, member D (CLEC2D) is implicated in the immune response. Sensing tissue damage is an ancient function of immune cells that is central to the regulation of inflammation, tissue repair, and immunity. IL-2RB is the subunit critical for receptor-mediated signaling via physically or functionally coupling to other signaling molecules, such as the Jak-STAT and Src-family protein tyrosine kinase although it lacks apparent catalytic motifs.

CD48 is a GPI-linked protein in the CD2 family of immunoglobulin superfamily proteins. CD48 is expressed on most lineage-committed hematopoietic cells but not on hematopoietic stem cells or multipotent hematopoietic progenitors. Among dendritic cells (DC), CD48 is selectively expressed on circulating myeloid DC and resident bone marrow and thymus DC. CD2, 2B4, and heparan sulfate function as CD48 ligands.
