MK-571 sodium
T3148115103-85-0
MK-571 sodium (L-660711 sodium salt) is a selective, orally active antagonist of the CysLT1 receptor. MK-571 sodium is a multidrug resistance protein-2 (ABCC2, Mrp2) inhibitor used to demonstrate the role of Mrp2 in the cellular efflux of drugs, xenobiotics, and their conjugates. MK-571 sodium can inhibit the synthesis of K-4′-O-GlcA (19.7 μM). MK571 dose-dependently inhibits the intracellular biosynthesis of all flavonol sulphates and glucuronides by Caco-2 cells. MK-571 sodium significantly inhibits phase-2 conjugation of kaempferol by cell-free extracts of Caco-2, and production of kaempferol-4′-O-glucuronide was competitively inhibited. In addition to inhibiting MRP2, MK571 is a potent inhibitor of enterocyte phase-2 conjugation.

Oroxylin A
T6S1315480-11-5
1. Oroxylin A (Baicalein 6-methyl ether) has various anti-tumor effects including apoptosis, cell cycle arrest, drug-resistant reversion. 2. Oroxylin A possesses abilities of inhibiting the ATRA-induced IL-6 production via modulation of LAP/LIP/CHOP in leukemia cell lines, which could providing a therapeutic strategy for RAS. 3. Oroxylin A inhibits UCP2s triggers the MPTP opening, and promotes the apoptosis in CaCo-2 cells; uncoupling protein 2 plays a key role in mitochondrial apoptotic pathway. 4. Oroxylin A inhibits N1ICD translocating to the nucleus and binding to epithelial-mesenchymal transition-related transcription factor Snail, thus suppressing the invasion and migration of MCF-7 cells. 5. Oroxylin A improves the sensitivity of K562/ADM cells by increasing apoptosis in leukemic cells and decreasing the expression of CXCR4 and PI3K/Akt/NF-κB pathway, and probably served as a most promising agent for CML treatment.

9(Z),11(E),13(E)-Octadecatrienoic Acid ethyl ester
T3688842021-86-3
9(Z),11(E),13(E)-Octadecatrienoic Acid ethyl ester (α-ESA) is a conjugated polyunsaturated fatty acid commonly found in plant seed oil. This fatty acid accounts for about 60% of the total fatty acid composition of bitter gourd seed oil and about 70% in tung oil. α-ESA is metabolized and converted to conjugated linoleic acid (9Z,11E-CLA) in rats. It has shown potential as a tumor growth suppressor. In colon cancer Caco-2 cells, α-ESA induced apoptosis through up-regulation of GADD45, p53, and PPARγ. In DLD-1 cells supplemented with α-ESA, apoptosis was induced via lipid peroxidation with an EC50 of 20 μM. It also inhibits DNA polymerases and topoisomerases with IC50s ranging from ~5-20 μM for different isoforms of the enzymes. α-ESA ethyl ester is a neutral, more lipid soluble form of the free acid.

Valganciclovir
T61259175865-60-8
Valganciclovir, the L-valyl ester of ganciclovir, is a prodrug effectively converted to ganciclovir in the body, serving as an antiviral medication for cytomegalovirus (CMV) infections. In vitro studies demonstrate that valganciclovir inhibits glycylsarcosine transport mediated by PEPT1 and PEPT2 in Caco-2 and SKPT cells, respectively, with competitive inhibition K(i) values of 1.68±0.30 mM for PEPT1 and 0.043±0.005 mM for PEPT2. Clinical trials indicate valganciclovir's non-inferior efficacy to ganciclovir in pre-emptive therapy for CMV, exhibiting no significant difference in viral clearance rates or toxicity levels between the treatments. Specifically, 89.5% of patients treated with valganciclovir and 83% treated with ganciclovir achieved viral clearance at 28 days (P=0.030 for non-inferiority). Additionally, valganciclovir prophylaxis at 450 mg twice daily prevented CMV reactivation in patients on an alemtuzumab-containing regimen, with none of the 20 patients experiencing reactivation (P=0.004).
